AI heeft het net een stuk makkelijker gemaakt.<\/p>\n<h2>Modellen voor diep leren<\/h2>\n<p>De uitdaging bij het maken van een nieuw antibioticum is dat er bijna oneindig veel moleculaire regelingen zijn en het is moeilijk om te weten welke daarvan een specifieke bacterie zal doden. Om het nog ingewikkelder te maken, moeten onderzoekers ervoor zorgen dat de nieuwe verbinding ook geen gezonde cellen doodt.<\/p>\n<p>Het MIT-team cre\u00eberde een database van 39.000 verschillende verbindingen en hun effecten op Staphylococcus aureus. Ze namen ook de cytotoxiciteit van de verbindingen op door de effecten te volgen die ze hebben op menselijke lever-, skeletspier- en longcellen.<\/p>\n<p>Informatie over de chemische structuren van de verbindingen werd ook toegevoegd aan de dataset, die vervolgens werd gebruikt om een deep learning AI-model te trainen.<\/p>\n<p>Op basis van de grote dataset kon het model leren welke chemische structuren het meest waarschijnlijk de bacteri\u00ebn zouden doden zonder nadelige effecten op gezonde cellen.<\/p>\n<p>Nadat het AI-model was getraind, werd het gebruikt om 12 miljoen commercieel beschikbare verbindingen te screenen. Het model identificeerde moleculen uit vijf verschillende klassen waarvan het voorspelde dat ze de MRSA-bacterie zouden doden.<\/p>\n<p>Op basis van deze sterk verkleinde lijst van kandidaat-verbindingen verkregen de onderzoekers 280 verbindingen om te testen tegen MRSA-bacteri\u00ebn die in een laboratoriumschaaltje werden gekweekt. Hun tests leidden tot de ontdekking dat twee van de verbindingen MRSA met een factor 10 verminderden.<\/p>\n<h2>Verklaarbare AI<\/h2>\n<p>AI-modellen geven ons vaak bruikbare antwoorden, maar het kunnen ondoorgrondelijke zwarte dozen zijn, die ons geen inzicht geven in hoe ze tot het antwoord komen.<\/p>\n<p>Felix Wong, een postdoc bij MIT en Harvard en samen met Erica Zheng een van de hoofdauteurs van het onderzoek, zei: \"Wat we met dit onderzoek wilden doen, was de zwarte doos openen.
