Het c<\/span><span style=\"font-weight: 400;\">brengt de tumor gedetailleerder in kaart en onthult cel-tot-cel interacties en hoe deze correleren met het resultaat voor de pati\u00ebnt.<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Gevaert: \"Het model liet zien welke cellen graag bij elkaar zijn, welke cellen niet met elkaar willen communiceren en hoe dit correleert met pati\u00ebntresultaten.\"\u00a0<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Het model ontdekte bijvoorbeeld dat de clustering van specifieke cellen, bekend als astrocyten, duidde op een agressievere vorm van de kanker. Deze inzichten kunnen mogelijk helpen bij het ontwerpen van effectievere behandelingen voor glioblastoom.<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Zheng hoopt dat het model ook kan dienen als hulpmiddel voor postoperatieve beoordeling. Het model toonde aan dat tumorcellen die tekenen van zuurstoftekort vertonen vaak samenhangen met slechtere kankerresultaten.\u00a0<\/span><\/p>\n<p><span style=\"font-weight: 400;\">\"Door de zuurstofarme cellen in histologisch gekleurde operatiemonsters te belichten, kan het model chirurgen helpen begrijpen hoeveel kankercellen er mogelijk nog in de hersenen aanwezig zijn en hoe snel de behandeling na de operatie moet worden hervat\", aldus Zheng.<\/span><\/p>\n<p><span style=\"font-weight: 400;\"> Hoewel het model zich nog in de onderzoeksfase bevindt, kan het ook worden toegepast op andere vormen van kanker, zoals borst- of longkanker.<\/span><\/p>\n<p><span style=\"font-weight: 400;\"> Zheng concludeerde: \"Ik denk dat deze multimodale data-integraties de verbetering van gepersonaliseerde geneeskunde in de toekomst vorm kunnen geven.\"<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Momenteel is er een proof-of-conceptversie van hun model, GBM360 genaamd, beschikbaar voor onderzoekers om diagnostische beelden te testen en te uploaden om de uitkomsten van glioblastoma-pati\u00ebnten te voorspellen. <\/span><\/p>\n<p><span style=\"font-weight: 400;\">Zheng haastte zich echter toe te voegen dat het model zich nog in de onderzoeksfase bevindt en nog niet in echte klinische omgevingen wordt gebruikt.\u00a0<\/span><\/p>\n<h2><span style=\"font-weight: 400;\">Meer over het onderzoek<\/span><\/h2>\n<p><span style=\"font-weight: 400;\">Het onderzoek maakt gebruik van AI om glioblastoma-subtypes te interpreteren op basis van bestaande pati\u00ebntgegevens, zodat clinici de prognose en ontwikkeling van de ziekte voor verschillende pati\u00ebnten kunnen bepalen.\u00a0<\/span><\/p>\n<p>Zo werkt het:<\/p>\n<ol>\n<li style=\"font-weight: 400;\" aria-level=\"1\"><span style=\"font-weight: 400;\"><strong>Gegevensintegratie<\/strong>: De onderzoekers begonnen met het integreren van meerdere vormen van data, waaronder single-cell RNA sequencing en spatial transcriptomics, naast klinische resultaten van glioblastoma pati\u00ebnten. Dit leverde een robuuste dataset op voor het ontwikkelen van machine learning (ML) modellen.<\/span><\/li>\n<li style=\"font-weight: 400;\" aria-level=\"1\"><span style=\"font-weight: 400;\"><strong>Modelontwikkeling<\/strong>: Het team ontwikkelde GBM-CNN, een gespecialiseerd deep-learning model dat is ontworpen om histologiebeelden te interpreteren. Dit model werd getraind om de verschillende transcriptionele subtypes van glioblastoomcellen te voorspellen, waarbij de ge\u00efntegreerde gegevens werden gebruikt voor validatie.<\/span><\/li>\n<li style=\"font-weight: 400;\" aria-level=\"1\"><span style=\"font-weight: 400;\"><strong>Grootschalige analyse<\/strong>: Na training werd GBM-CNN gebruikt om meer dan 40 miljoen weefselspots van 410 pati\u00ebnten te analyseren, waardoor cellulaire kaarten met een hoge resolutie werden gemaakt. De analyse onthulde de aanwezigheid van drie tot vijf kwaadaardige subtypen in elke tumor.<\/span><\/li>\n<li style=\"font-weight: 400;\" aria-level=\"1\"><span style=\"font-weight: 400;\"><strong>Correlatie met klinische gegevens<\/strong>: De cellulaire kaarten werden vervolgens ge\u00efntegreerd met klinische gegevens van pati\u00ebnten. Er werd ontdekt dat bepaalde cellulaire composities verband houden met slechtere pati\u00ebntresultaten.<\/span><\/li>\n<li style=\"font-weight: 400;\" aria-level=\"1\"><span style=\"font-weight: 400;\"><strong>Validatie en testen<\/strong>: Om deze bevindingen te bevestigen werd een secundair model ontwikkeld om de prognose van pati\u00ebnten te voorspellen op basis van andere diagnostische beelden.
