Deze geneesmiddelen werken door senescente cellen, vaak 'zombiecellen' genoemd, te elimineren. Zulke cellen blijven metabolisch actief maar kunnen zich niet vermenigvuldigen.<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Hoewel het stoppen van celreplicatie werkt als een defensief mechanisme om de verspreiding van celschade te voorkomen, is het niet helemaal eenvoudig.\u00a0<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Deze beschermende maatregel is vooral bedoeld om de proliferatie te voorkomen van cellen die DNA-schade hebben opgelopen - bijvoorbeeld cellen die zijn beschadigd door blootstelling aan zonlicht. Door de replicatie van deze beschadigde cellen te stoppen, zorgt het lichaam ervoor dat deze schade zich niet naar meer cellen verspreidt.<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Deze cellen die gestopt zijn met delen, bekend als senescente cellen, kunnen echter mogelijk schade veroorzaken. Wanneer ze in de senescentiefase komen, beginnen ze eiwitten af te geven waarvan bekend is dat ze ontstekingen veroorzaken.\u00a0<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Dit kan naburige gezonde cellen aantasten en mogelijk een ontstekingsomgeving cre\u00ebren.\u00a0<\/span><span style=\"font-weight: 400;\">Na verloop van tijd kan deze opeenstapeling van ontstekingen bijdragen aan verschillende gezondheidscomplicaties.\u00a0<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Zo is ontsteking gekoppeld aan veel ziekten, waaronder neurologische en degeneratieve ziekten, diabetes type 2, longfibrose, artrose en kanker.\u00a0<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Hoewel het stoppen van de celreplicatie dus cruciaal is om de verspreiding van DNA-schade te voorkomen, kan de ontstekingsreactie die gepaard gaat met senescente cellen leiden tot andere gezondheidsproblemen als er niet goed mee wordt omgegaan.<\/span><\/p>\n<p><span style=\"font-weight: 400;\">En dat is waar senolytische medicijnen kunnen helpen.\u00a0<\/span><\/p>\n<h2><span style=\"font-weight: 400;\">De rol van AI<\/span><\/h2>\n<p><span style=\"font-weight: 400;\">Eerder onderzoek op laboratoriummuizen toonde aan dat senolytische medicijnen dit ontstekingsscenario verbeteren door senescente cellen te elimineren en gezonde cellen te sparen.\u00a0<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Hoewel er ongeveer 80 bekende senolytica bestaan, is slechts een combinatie van twee daarvan, dasatinib en quercetine, getest bij mensen.\u00a0<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Het ontdekken van geneesmiddelen is uitzonderlijk duur en duurt meestal 10 tot 20 jaar - het is een belangrijk knelpunt in de ontwikkelingspijplijn van geneesmiddelen. In dit onderzoek gebruikten onderzoekers machine learning (ML) om de ontdekking van nieuwe senolytische geneesmiddelen te versnellen.\u00a0<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Ze trainden AI-modellen op bestaande senolytica en niet-senolytica, waardoor de AI onderscheid kon maken tussen deze twee en de waarschijnlijkheid kon voorspellen dat niet-ontdekte moleculen senolytica waren.<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Van de 4.340 ge\u00ebvalueerde moleculen markeerde de AI er 21 met een hoge waarschijnlijkheid binnen 5 minuten als senolytisch.\u00a0<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Een van de auteurs van het onderzoek, Dr. Vanessa Smer-Barreto, <\/span><a href=\"https:\/\/theconversation.com\/ai-finds-drugs-that-could-fight-ageing-and-age-related-diseases-208390#:~:text=The%20results%20showed%20that%20out,they%20work%20in%20the%20body.\"><span style=\"font-weight: 400;\">vertelde The Conversation<\/span><\/a><span style=\"font-weight: 400;\"> dat, als het op traditionele wijze in een laboratorium getest zou worden, het alleen al om de aankoop van de verbindingen 50.000 pond zou kosten, exclusief de kosten van apparatuur en installatie.<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Na in vitro testen op gezonde en senescente cellen bleken 3 van de 21 moleculen, periplocin, oleandrin en Linkedin, effectief senescente cellen te elimineren en normale cellen te sparen.\u00a0<\/span><\/p>\n<p><span style=\"font-weight: 400;\">In verdere tests presteerde oleandrine, een verbinding die wordt gevonden in de oleanderplant, beter dan de bekendste senolytica in zijn soort.<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Het team test nu de 3 kandidaat-senolytica op menselijk longweefsel en de resultaten worden over ongeveer twee jaar verwacht.<\/span><\/p>\n<p><span style=\"font-weight: 400;\">In de afgelopen maanden heeft AI <\/span><a href=\"https:\/\/dailyai.com\/nl\/2023\/05\/ai-helps-researchers-discover-a-new-antibiotic-for-treating-drug-resistant-bacteria\/\"><span style=\"font-weight: 400;\">ge\u00efdentificeerde potenti\u00eble antibiotica<\/span><\/a><span style=\"font-weight: 400;\"> effectief is in het doden van bacteri\u00ebn die resistent zijn tegen medicijnen, en een biotechbedrijf uit Hong Kong kreeg goedkeuring om klinische tests te beginnen met een <\/span><a href=\"https:\/\/dailyai.com\/nl\/2023\/07\/first-ai-generated-drug-given-go-ahead-for-human-trials\/\"><span style=\"font-weight: 400;\">AI-ontdekt geneesmiddel<\/span><\/a><span style=\"font-weight: 400;\">.\u00a0<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Het vermogen van AI om complexe analyses van moleculen uit te voeren, versnelt de ontwikkeling van farmaceutische pijplijnen enorm en biedt nieuwe mogelijkheden om veelvoorkomende en moeilijk te behandelen aandoeningen en ziekten aan te pakken. <\/span><\/p>","protected":false},"excerpt":{"rendered":"<p>Een internationaal team van onderzoekers van de Universiteit van Edinburgh en de Spaanse Nationale Onderzoeksraad IBBTEC-CSIC gebruikten AI om de ontdekking van geneesmiddelen tegen veroudering te versnellen.
